We researched further into restriction enzymes, looking for a known cut site and its restriction enzyme, so that in our project we can methylate that site and know it was succesfully methylated, because the restriction enzymes won't cut if there's a methyl group. We also began to contact the iGEM Heidelberg team (2014) to get more specifics on their iGEM project (preserving methylation in PCR). We also attempted to get in contact with New England Bio Labs, in order to talk to them about chemical PCR, such as with a salt.
