Today we finalized our timeline and contacted the company Qiagen about a kit they have for "methylation specific PCR (MSP)." Our plan is to, by tomorrow's class, have finished research on helicase dependent PCR and MSP, in order to decide which (or both) form of PCR to use.

 

TIMELINE:

Week of February 15

  • Feb 15: Finish choosing PCR Type

  • Feb 17: Continue research on chosen PCR Type

  • Feb 18: (Finish?) research on PCR Type, begin designing DNA

Week of February 22 - NINTH WEEK

  • Feb 22: begin work on poster this week (?), do any further research from last week

Week of February 29

  • Feb 29: Finish designing DNA, begin working on control for assay (Lab work/research)

  • March 2: Continue working on control for assay (Lab work)

  • March 3: Finish control for assay work, begin tinkering with protocols (Lab work/research)

Week of March 7

  • March 7: Continue tinkering with protocols, begin assay testing

  • March 9: Continue testing assay, tinkering with protocols

  • March 10: Continue testing assay, tinkering with protocols

Week of March 14

  • March 14: Finish all lab work, finish poster

  • March 16: Close any loose ends

  • March 17: Complete all goals (see below and flowchart)

  • perform PCR on methylated DNA and detect the methylation patterns; Will the DNA remain methylated after PCR?

  • have done electrophoresis to prove DNA is methylated
