Today we were a bit scattered. We had to remind ourselves of what had happened in the past few weeks. After some confusion, we began drafting questions for Anna Huhn, who worked with the Heidelburg iGEM team in 2014. Hopefully she can answer some of our pressing questions. We also tried to contact the UVA iGEM team; in the hopes that they can provide us with DNA from the 2015 distribution kit they were given by iGEM. Our next step is to design our DNA and get it built, so that we can design primers and begin experimentation.

Questions for Anna Huhn:

  1. How did you design and then obtain your DNA?

  2. How did you test for methylation? Did you get any PCR results?

  3. What buffers did you use for PCR2.0?

  4. Do you need other proteins to help DNMT1 attach to the DNA?

  5. If PCR2.0 is circular, how does it attach to the DNA?
